I have a 12mm plywood sheet with 24mm (2 x 12mm) plywood pieces glued around the edges using Titebond III. This item is going to get banged around, loaded in and out of cars, etc. Should I be concerned that the glue alone won't be enough to keep it together?

I have some wood screws but I understand that screwing into the plywood edges (via the large sheet) would be a bad idea without some fussing with CA glue or other methods to keep the plies together in the long term.

I thought of gluing blocks of wood into the corners then one screw in each direction (side, side, sheet). Would that make much of a difference?

Any other suggestions?

For additional strength I’d use your corner block suggestion . The screws act as clamps while the glue sets and will give the plywood extra strength especially if it going to get knocked or dropped . You could also use pocket screws but I think the corner blocks and glue is a better plan ..
 
Sorry too late now but the two layers glued together should have been made to form Lap joints at the corners.
You can screw into the edge but very carefully after drilling to core of thread size.

Pre drilling will minimise risk of splitting the lamination, also blunt the end of your screw before using it.
