I would use glue direct from the bottle (1 litre sized) and just run a bead down the centre of the board edge. Let the compression of the components spread the glue out for you. Once assembled have a quick nose round and clean up any squeeze out.

FWIW.

As above some 25mm brads or staples fired in at an angle would prolly do the job for the back once the glue is set.
 
I'd glue the back on and secure with Brads. You're in production line mode so a brad nailer would hold the back in place while the glue dries and you'll be able to fix the back in place very quickly. The back will provide torsional strength but it's not going to have weight applied that would force the brads out, so that would be my approach. Get a decent glue bottle with a roller as well so you can quickly run the glue around the frame.
