Is it worth me approaching some manufacturers (e.g. medite) for 20 sheets of mdf or would that amount be too small for them to bother with?

Ask your local suppliers if they'll give a discount/cash. Why not? Tbh 20 is nowt to a builders merchants. Might get a couple of quid off. It's possible... I think wholesale is pushing it lol. Anyone you know have a trade account?

Find out how many is on a normal full pallet, that will be the best price break. They wont need to proccess it at all just deliver it to you.
Try importers like severn ply or whatever they are called now, Timbmet, Arnold lavers, Avon plywood etc

Many moons ago when I was starting out, I contacted the manufacturer to try to negotiate a advantageous price. Every time I called, "He wasn't in the office"

So I recalled a tip I read somewhere that the best time to get the boss was during lunch hour, as everyone else was out.

I phoned at 13:30 mid week and he answered the phone.

Alas it did no good. he said he couldn't sell to me even if I would take a lorry load.

But he did put me in touch with the main distributor who treated me very well.

A mate buys 18mm MDF by the pack which is 50 sheets to get a better price & that’s still not from the manufacturer. I can’t see a manufacturer being interested in supplying anything less than a lorry load & would expect you to have the machinery to unload it.
